Default Pouring the suspended slab and core fillinh at the same time?

Without a cement mixer im going to get a truck of premix in. I am considering building the base with hollow concret blocks then framing the suspended slab and pouring that as well as filling the cores at the same time. Has this been done? I was thinking i could use a construction glue to hold the blocks in place for the pour.

You can do it in one pour. In fact this will be slightly stronger that two separate pours.

If you are careful during the pour, the blocks should hold together well enough by gravity. If you want to be sure, mortar the blocks. The mortar is probably cheaper than construction adhesive.
